RRB Paramedical Lab Technician Syllabus

The RRB Paramedical exam for the post of Lab Technician (Laboratory Assistant Grade II) is a single-stage CBT of 100 marks in 90 minutes. The 70-mark Professional Ability section tests your knowledge of medical laboratory science from your DMLT or B.Sc MLT curriculum. Professional Ability Syllabus for Lab Technician (70 Marks)


Haematology and Blood Banking

One of the most important topics in the Lab Technician paper. Covers the collection, processing, and analysis of blood samples, complete blood count (CBC) and its interpretation, haemoglobin estimation methods (Sahli, Cyanmethaemoglobin), ESR methods (Westergren, Wintrobe), blood grouping and cross-matching, coagulation tests (PT, APTT, BT, CT), and basics of blood banking including blood components and their storage conditions.


Clinical Biochemistry

Covers laboratory analysis of blood and urine for biochemical parameters: blood glucose (fasting, post-prandial, HbA1c), kidney function tests (urea, creatinine, uric acid), liver function tests (bilirubin, ALT, AST, ALP, total protein), lipid profile, thyroid function tests, and cardiac markers. Know the normal reference ranges for all major parameters.


Microbiology

Covers culture and sensitivity techniques, staining methods (Gram stain, Ziehl-Neelsen stain, Giemsa stain), sterilisation and disinfection methods, identification of common pathogenic bacteria and fungi, parasitology (identification of common intestinal and blood parasites), and basics of virology. Know the agar types (blood agar, MacConkey agar, CLED agar) and their uses.


Histopathology and Cytology

Covers tissue processing, fixation, embedding, and sectioning, H&E staining procedure, special stains and their purposes (PAS, Masson Trichrome, AFB stain), cytological examination of body fluids, FNAC (Fine Needle Aspiration Cytology), and PAP smear preparation and interpretation. Know the WHO classification of common tumours at a basic level.


Clinical Pathology and Urine Analysis

Covers routine and microscopic examination of urine (colour, transparency, pH, specific gravity, protein, glucose, ketones, urobilinogen, bile salts and pigments), urine microscopy for cells and casts, examination of other body fluids (CSF, pleural fluid, synovial fluid, semen analysis), and stool examination for ova, cysts, and occult blood.


Immunology and Serology

Covers antigen-antibody reactions and their applications in laboratory diagnosis: ELISA, WIDAL test, VDRL/RPR (syphilis serology), ASO titre, RA factor, HIV testing algorithm, hepatitis B markers (HBsAg), and Dengue NS1 antigen and antibody tests. Understand the principle behind each serological test.


Laboratory Management and Safety

Covers biomedical waste management rules (colour-coded waste segregation), laboratory safety practices, infection control in the lab, quality control concepts (internal and external QC), calibration of lab instruments, and basic principles of major instruments like spectrophotometer, centrifuge, haematology analyser, and ABG analyser.
